Lysinibacillus sphaericus es una bacteria gram positiva ampliamente estudiada por sus propiedades entomopatógenas y su tolerancia a diferentes metales pesados. Sin embargo, debido a su alta conservación genómica y a la ardua transferencia horizontal de ADN foráneo, la manipulación genética de cepas de L. sphaericus con características ambientales importantes se ha ralentizado. Recientemente, se observó la presencia de diferentes sistemas de restricción-metilación en la cepa entomopatógena L. sphaericus C3-4, los cuales estarían involucrados en la regulación del material genético externo que entra a la célula y podrían actuar como una barrera que garantiza la identidad bacteriana. Ya que existe alta similaridad evolutiva entre cepas tóxicas, este estudio evaluó los sistemas de restricción-metilación de la cepa colombiana entomopatógena L. sphaericus III (3) 7 como una ventaja en su transformación con el plásmido pMK4-ampR / chlR. Resumen
Lysinibacillus sphaericus is a gram positive bacteria widely studied for its entomopathogenic properties and its tolerance to different heavy metals. However, due to its high genomic conservation and to the arduous horizontal transfer of foreign DNA, genetic manipulation of L. sphaericus strains with significant environmental features has been slowed. Recently, the presence of different restriction-methylation systems was observed in the entomopathogenic strain L. sphaericus C3-4, those would be involved in the regulation of external genetic material into the cell and could act as a barrier that ensures bacterial identity. Since there is a high evolutionary relatedness between toxic strains, this study assessed the restriction-methylation systems of the entomopathogenic Colombian strain L. sphaericus III(3)7 as an advantage in its transformation with the plasmid pMK4-ampR/chlR.
