    Cuantificación de la expresión del factor de transcripción Foxp3 en pacientes diagnosticados con neuromielitis óptica (NMOSD) y un grupo de controles de la ciudad de Bogotá-Colombia: una revisión de su función en autoinmunidad

    Objetive: Regulatory T cells (Tregs) are critical mediators of central tolerance, this cell population are defined by the expression of Foxp3 (Forkhead box p3) a transcription factor that plays a critical role in their immunological function, development and maintenance. The aim of this study was to quantify Foxp3 mRNA expression level in mononuclear cells from periferal blood samples (PBMC) of a group of participants diagnosed with neuromyelitis optica NMOSD and healthy controls from Bogotá-Colombia as an approach to transcriptional activity comparison and validation in the context of this disease. Methods: RNA was extracted from PBMC obtained from whole blood samples of NMO participants and healthy controls. RT-qPCR analysis was used to evaluate FoxP3 mRNA expression.β-Actin gene was used as the reaction normalizer. The 2-^^Ct method was used as the relative quantification strategy for RT-qPCR data analysis. Results: A total of 26 participants (42.42 mean age; 76.92% female) and 15 controls (30.27 mean age; 66.6% female) were included in the analysis. Foxp3 mRNA mean expression level was significantly different between NMO diagnosed participants and controls (p<0.05). Conclusions: The relative quantification of Foxp3 transcription level through RT-qPCR is considered as the primary approach to the triggers and complex underlying mechanisms that are involved in the loss of tolerance generation of autoimmunity in NMO and along the autoimmune spectrum. It is relevant to continue monitoring the activity of this transcription factor in the context and along the NMO spectrum disease.

    Analysis of gene expression profiles related to phenol degradation in the environmental strain Scedosporium apiospermum HDO1

    Filamentous fungi are efficient microorganisms in the degradation of petroleum hydrocarbons. Among them Scedosporium apiospermum has been reported as able to degrade phenol, a pollutant widely produced in industrial waste. Two initial phenol degradation pathways have been proposed for the fungal species: one via catechol and the other through hydroquinone. Based on genomic information, genes related to the latter two pathways coding for phenol hydroxylase and catechol 1,2-dioxygenase have been reported in the environmental strain S.apiospermum HDO1. The present study assessed the transcriptional changes of the genes involved in the phenol degradation pathways proposed in S.apiospermum HDO1. The fungal strain was grown during four days in Salts Minimum Medium under two carbon sources, phenol or glucose (control). The mycelium grown was collected and sent for RNA extraction and then sequencing with RNA-seq (Illumina Hiseq-4000 platform). For the five enzymes previously reported for the ortho-catechol pathway, we identified four genes codifying for phenol 2-monooxygenase, three genes for Catechol dioxygenase, and one gene codifying for hydroxyquinol 1,2-dioxygenase. Several genes involved in the first steps of the pollutant transformation were differentially expressed, these results suggest that the degradation of phenol by S.apiospermum is achieved via the ortho-catechol pathway.

    Effect of the absence of the transcription factor hand2 on the fibronectin expression during Jaw Development of Zebrafish (Danio rerio)

    In jaw development, various transcription factors contribute to the definition of cell fate and the organization of the extracellular matrix. One of these transcription factors is Hand2. Its absence in other tissue contexts has shown to affect fibronectin expression and distribution, an essential component of the extracellular matrix. In fact, previous studies have shown that the absence of the transcription factor Hand2 leads to fibronectin overexpression in the heart. These changes could also affect jaw development in zebrafish (Danio rerio). However, it is not known yet if there is any effect of fibronectin misexpression during jaw development. Therefore, we are evaluating how the absence of the transcription factor Hand2 changes the fibronectin expression and distribution in the context of jaw development. Thus, we evaluate fibronectin expression in zebrafish at stages 24hpf, 36hpf, 48hpf, 72hpf by RT-PCR. We are also evaluating through in situ hybridization other markers like pitx2 and notch2 to identify the expression patterns in hand2 loss-of-function mutant embryos and their wild-type siblings. Our results will help us to understand some of the causes of mandibular defects such as those produced in the Apert and Sturge- Weber syndromes caused by alterations in the expression of hand2 in its role regulating fibronectin protein expression.

    Following a trail of biological collections records, historical distributions, and environmental DNA to find the lost harlequin toads of Colombia

    Neotropical harlequin toads (Anura: Bufonidae: Atelopus) face a severe threat, with 83% of the 94 assessed species classified as critically endangered or extinct by the IUCN. Their populations have experienced a significant decline, primarily due to emerging infectious diseases, even in pristine environments. In Colombia, where 41 native species exist, most have not been sighted in over three decades, raising concerns about potential extinctions. To enhance the chances of locating these elusive amphibians, we utilized data from biological collections, identifying key variables such as precipitation, month, stages, and sex that explain SVL length throughout the year. This information guides to the optimal time and location for search efforts. Additionally, species distribution models (SDMs) were employed in conjunction with environmental DNA (eDNA) analysis. SDMs, utilizing presence data from eDNA samples, are enhanced by incorporating environmental variables, facilitating a comprehensive understanding of the habitat requirements of each species, which contributes to targeted conservation efforts. The combination of eDNA and SDMs directs conservation projects towards effective strategies for the protection of Atelopus. Our study focused on sites with stable Atelopus populations of three species, two high mountains and one lowland, which served as a proof of concept, and two sites with severely depleted but persistent populations in the highlands. As we navigate the imperative task of safeguarding these frogs from extinction, the integration of innovative tools like eDNA and SDMs becomes indispensable. These findings are instrumental in shaping future conservation initiatives for this endangered group.

    Morphological and phylogenetic study reveals the presence of the three major clades of the genus Morchella in Colombia

    Taxonomic rearrangements have occurred in the Morchella genus in the past decade with the implementation of molecular tools. In this study, the diversity of these highly prized edible mushrooms in Colombia was evaluated with an integrative approach including morphological, ecological and molecular data (portions of the genes for RNA polymerase II largest subunit (RPB1) and second largest subunit (RPB2), translation elongation factor-1a (TEF1), the nuc rDNA region encompassing the internal transcribed spacers 1 and 2, along with the 5.8S rDNA (ITS)). Results showed, the presence in 12 departments of seven species in the three major clades of the genus. Two new species were found Morchella puracensis nom. prov. In the Esculenta clade, and MelNN from Pauna Boyacá in the Elata clade. Some species found present continentalism such as Morchella gracillis/Mes-14, and other cosmopolitan distribution like M. rufobrunnea and M. galilaeae/Mes-16 and a new variety of M. tridentina/Mel-2 from southern Colombia and Perú is proposed. This study assessed the ecology, distribution, and diversity of this natural resource with perspectives for its conservation and a sustainable use of morels from the Neotropics.

    The reproductive biology of the glassfrog centrolene solitaria: breeding behavior, advertisement call, and tadpole morphology

    The present study, focused in studying aspects related to the reproductive biology of the glassfrog Centrolene solitaria, a species that lacks associated natural history information. Between March and October 2018, we studied a population of C. solitaria in a locality in the department of Huila, Colombia. We conducted nocturnal and diurnal surveys, we calculated mating rates and parental care for males, examined phenotypic correlates of mating success, and described some aspects of the natural history of the species. We found that the mating success of the males increased with the number of nights they remain active, but not with the body size or the calling-perch height. Parental care was carried out exclusively by males, and the period of attention to the eggs is extended. The egg positions, deposited in riparian vegetation, consist of an average of 15 eggs (12-23), located on the upper part of the leaves between 0.6 - 3.0 m in height. The advertisement call of C. solitaria consists of three to four notes pulsed at an average frequency of 6298.67 Hz (6201.3-6378.8 Hz); the mean note duration was 0.036 s (0.031-0.045 s), and the mean pulse duration was 0.017 s (0.011-0.032 s). The tadpole is fosorial, slightly pigmented, with an elliptical body from the lateral view, and an anteroventral mouth with a dental formula LTRF: 2 (2) / 3 (1).

    Biología reproductiva de tres coníferas colombianas de la región Andina: Podocarpus oleifolius, Prumnopitys montana y Retrophyllum rospigliosii (Podocarpaceae)

    El ciclo reproductivo de las plantas es fundamental para comprender aspectos evolutivos, de diversificación y el estado ecológico actual de las especies. Una aproximación desde el estudio morfológico y anatómico de las estructuras reproductivas y los patrones fenológicos en especies poco estudiadas en Colombia como los son las podocarpáceaes andinas: Podocarpus oleifolius, Prumnopitys montana y Retrophyllum rospigliosii, aporta información a nuestro conocimiento del comportamiento reproductivo en especies tropicales de zonas montañosas, además puede sentar las bases para la toma decisiones en cuanto a conservación, especialmente en el contexto de hábitats vulnerables como los bosques andinos y cambio climático por causas antropogénicas. Se encontró un patrón episódico y sincronía en la producción de las estructuras reproductivas, con una clara tendencia la bimodalidad. Se observaron ciclos reproductivos prolongados de 2 años para la especiea de Podocarpus, 2 y 4 meses para la de Retrophyllum y 2 años y 7 meses para la de Prumnopitys. Además, se examinó la posible influencia de factores abióticos tales como la precipitación, el control fotoperiódico y la temperatura sobre la producción de óvulos, polen y semillas maduras. Los resultados muestran una fuerte relación entre varios factores abióticos y los eventos reproductivos. Para la liberación del polen son importantes los periodos de baja precipitación, para la aparición de óvulos tanto la precipitación como la insolación y la temperatura tienen un papel relevante y para la maduración de la semilla es la insolación quien esta mayormente asociada al suceso.

    Evaluación del daño en el ADN en niños menores de 5 años en dos zonas distintas de contaminación ambiental en la ciudad de Bogotá

    La contaminación del aire en las ciudades es una problemática a nivel mundial debido al gran impacto que tiene en la salud humana, y uno de los factores más relevantes son las emisiones de material particulado (PM). Los niños son más susceptibles debido a que sus pulmones, cerebro y sistema inmune se encuentran en desarrollo. La exposición a PM, especialmente a PM2.5, aumenta los niveles y la acumulación de especies reactivas de oxígeno (ROS), lo que promueve la respuesta al estrés oxidativo, y resulta en daño en el ADN. En este estudio, se evalúa la relación de la exposición reciente a contaminación ambiental, en una zona de alta y baja exposición a la contaminación, sobre la salud a través del uso de biomarcadores de exposición y daño en el ADN, en 169 niños menores de 5 años en la ciudad de Bogotá, Colombia; esto por medio del ensayo cometa y la determinación de polimorfismos relacionados con genes de enzimas de metabolismo (CYP1A1, GSTT1 y GSTM1), respuesta antioxidante (SOD2 y CAT) y reparación de ADN (OGG1 y p53BP1). Los resultados evidenciaron diferencias entre las zonas de alta y baja exposición con relación al daño en el ADN, diferencias significativas con relación al daño en el ADN y tres de los factores de susceptibilidad (GSTT1, SOD2 y CAT) y evidenciar factores de riesgo que aumentan las probabilidades de presentar un mayor daño del ADN relacionados con exposición a contaminantes intramurales. Es necesario aumentar las investigaciones con relación al daño que presenta este contaminante en la salud de los niños y poder comenzar a tomar medidas preventivas para la mitigación y el aumenta de enfermedades respiratorias en esta población.

    A Coffee Hope Tale: Coffea arabica Castillo and Caturra varieties survive in a drier world

    Colombia is one of the world´s largest coffee (Coffea sp.) producers and this product generates 2 million jobs approximately in the country. However, climate change threatens coffee cultivation in Colombia because predictions indicate that by 2050, some regions especially in the north, could face higher temperatures and reduced rainfall, having a negative impact on specialty coffee production. Consequently, understanding the resilience of different coffee varieties to drought becomes vital to increase the sustainability of Colombia´s coffee production. We investigated the thermotolerance and physiological responses to water deficit of two Coffea arabica varieties: Castillo and Caturra. Thermotolerance was assessed by determining the temperature that caused a 50 % reduction in quantum efficiency (Fv/Fm), which is an indicative of damage to photosystem II (T50). Castillo and Caturra exhibited similar T50 values (52.6 °C and 52.5 °C, respectively), demonstrating that both have strong resistance to thermal stress. To understand the physiological response and recovery from water shortage of Castillo and Caturra, we subjected 18 plants of each variety to a 27-day drought treatment in a growth chamber at 23/19 °C (day/night) with a control group continuously irrigated. We measured leaf water potential (Ψ), the osmotic potential at turgor loss point (Ψπtlp), stomatal conductance (g), transpiration (E), photosynthetic rates (A), and relative growth rates (RGR). Overall, Caturra was slightly more responsive to drought than Castillo because during the drought period, Castillo consistently exhibited higher Ψ, E, and A rates than Caturra, which showed reduced RGR. Conversely, under drought, Caturra showed lower Ψπtlp, suggesting a potential active osmotic adjustment. Upon rehydration, both varieties recovered to nearly the same physiological values as the control plants. Our results emphasize the responses to drought of Castillo and Caturra and their ability to recover following a 27-day water deficit. Future studies should explore the response of mature plants and the drought´s effect on fruit production to ascertain the optimal variety for cultivation in Colombia´s northern regions.

    Fixing the tree: a phylogenomic approach reveals a robust time scale phylogeny of the Terminal Fusarium Clade

    The Terminal Fusarium Clade (TFC) is a group of fungi in the family Nectriaceae with agricultural and clinical relevance. In recent years different phylogenies have been presented in the literature with disagreement in the topologies. Few studies analyzing the divergence time scale of the group have been done. The evolutionary history of the group is still uncertain. Therefore, this study aims to understand the evolutionary history of the TFC. To pursue this objective, we performed a phylogenomic analysis using available genomes in GenBank and eight different pipelines. Then we analyze the discordance between genes and species history. Finally, a time tree was inferred with the Bayesian approach. Here, we present a new robust topology of the TFC that differs in some nodes of previous studies. Our results suggest the need for phylogenomic analyses to obtain a confident topology. The discordance between the gene and species history could be related to insufficient data on a single gene. Also, we present a time tree that shows new times divergence differing from previous studies because of the topology discordances. In conclusion, we present a robust time scale phylogeny that allows us to formulate new hypotheses of the evolutionary history of the TFC that should be validated in future studies.

    Evaluation of satanin 1 as a potential antifungal drug to treat Malassezia infections

    Malassezia is a fungal genus that is part of the skin's normal mycobiota but can also cause various diseases. The emergence of resistance to antimicrobial agents in several microorganisms, including Malassezia species, has led to the exploration of new therapeutic alternatives such as antimicrobial peptides. This study aimed to investigate the effect of satanin 1, a recently identified antimicrobial peptide, against Malassezia using broth microdilution assays, SEM microscopy, and a Galleria mellonella infection model. Results showed that the MIC of satanin 1 against Malassezia ranged from 50 to 12.5 µg/mL and it works by affecting the fungal cell surface. Nonetheless, satanin 1 did not improve the survival of G. mellonella in the infection model, possibly due to the low stability of the peptide in the larvae; however, this might be addressed by modifying the peptide's structure to enhance its stability and efficacy. Thus, continued investigation into new alternative therapies, like antimicrobial peptides, is crucial to combat the increasing threat of antifungal resistant species.

    Evaluación del uso de bacteriófagos para el biocontrol de aislamientos de Klebsiella pneumoniae multidrogoresistente en cuatro materiales de superficies comunes en entornos clínicos

    La propagación de Infecciones Asociadas a la Atención en Salud (IAAS), principalmente por contaminación bacteriana, representa una seria amenaza para los sistemas de salud. La prevalencia de K. pneumoniae en hisopos de superficie, su detección en distintos materiales, la creciente resistencia global a carbapenémicos con tasas de mortalidad de 22-72% y cepas resistentes a desinfectantes, acentúan la necesidad de opciones biotecnológicas. Con el fin de explorar una alternativa de descontaminación efectiva y específica para el control de aislamientos de K. pneumoniae multirresistente, este estudio investigó el potencial de los fagos para controlar la carga bacteriana en materiales de superficies comunes en entornos clínicos: acero, tela, PVC y poliestireno. Se determinó que los aislamientos de K. pneumoniae sobrevivieron hasta 24 horas en superficies, con reducciones entre 0.49-1.59 y 1.69-3.88 LOG UFC a 24°C y 37°C, respectivamente. Posteriormente, se seleccionaron tres bacteriófagos de aguas residuales hospitalarias, que en conjunto mostraron actividad lítica contra el 88% de los aislamientos hospitalarios, además, de actividad antimicrobiana en superficies, logrando reducciones de hasta 4.99 LOG UFC. Estos resultados respaldan la efectividad de los fagos como desinfectantes y sugieren su potencial uso complementario en situaciones de brote en entornos hospitalarios.

    Phylogeography of a sex-limited polymorphism: Disentangling phenotypic variation in the Tourmaline Sunangel, Heliangelus exortis

    While much research has focused on the evolution of ornaments in male animals, females can also exhibit ornamental phenotypes and these have received much less attention. In the hummingbird species H. exortis (Tourmaline Sunangel), females appear to exhibit an intriguing pattern of variation across their geographic range in the tropical Andes, with some populations exhibiting a sex-limited polymorphism where females can have either a drab or variable male-like phenotype depending on the amount of iridescence on their gorgets. We conducted a phenotypic description of female plumage using pattern-based and reflectance spectrophotometry analyses across the distribution of H. exortis. We also performed a phylogeographic analysis to investigate how phenotypic variation relates to evolutionary relationships between populations and to gain insights into the origin and maintenance of the polymorphism. Our findings indicate that, in contrast to previous findings which suggested that geographic variation in the polymorphism was clinal, females in all Colombian populations exhibit a continuous range of polymorphic states, whereas those in Ecuador do not exhibit the polymorphism at all. Molecular analyses further revealed genetic differentiation among populations across the species distribution, highlighting a long history of isolation between populations and the role of inter-Andean valleys as major biogeographic barriers to gene flow. Additionally, our study uncovered a complex evolutionary history within the H. exortis clade, including an apparent loss of the polymorphism in the Ecuadorian population and the origin of a phenotypically distinct species, H. micraster, which is nested within H. exortis and sister to Ecuadorian populations. We propose that the phenotypic variation observed in H. exortis may be the result of genetic drift under geographic isolation or social selection acting differently within populations, leading to the maintenance of the female-limited polymorphism in Colombia and its loss in Ecuador. Our results emphasize the role that females can play in shaping the phenotypic and genetic diversity of species. We suggest that further molecular and socioecological studies are needed to better understand the evolution of the polymorphism and to delimit species boundaries within the genus.

    Aspectos genéticos de la adaptación a la altitud de la Cordillera de los Andes en poblaciones nativas del norte de Argentina

    En el presente estudio se identificaron y analizaron variantes genéticas de adaptación a la altitud en poblaciones de estudio nativas del norte Argentina en la cordillera de los Andes. El análisis se basó en determinar la presencia de variantes asociadas la altitud reportadas en literatura [17], mediante la genotificación de variantes en los genes reportados SFTPD, FAM213A y ANXA11 en dos poblaciones del norte de Argentina ubicadas en diferentes altitudes, se determinó la ancestría y la estructura genética poblacional y se realizó una recopilación de información de variantes asociadas a altitud reportadas de bases de datos. Todos estos datos en conjunto, permitieron contribuir al entendimiento del proceso de adaptación a la altitud en poblaciones de la cordillera de los andes, entendido más como un proceso hereditario de las dinámicas migratorias que un evento de adaptación independiente. Se propone ampliar el rango de variantes analizadas, así como la evaluación de aspectos epigenéticos y regulatorios asociados a la presencia de las mismas para determinar su roll especifico en el proceso adaptativo a la altitud de la cordillera de los Andes.

    Validación de la interacción entre el efector XopAE de Xanthomonas phaseoli pv. manihotis y una patelina de plantas involucrada en inmunidad vegetal

    El añublo bacteriano de la yuca (CBB, por sus siglas en inglés), causado por Xanthomonas phaseoli pv. manihotis (Xpm), es la enfermedad bacteriana más importante de la yuca y afecta la producción de este cultivo en varios países tropicales en todo el mundo. Al igual que otras Xanthomonas fitopatógenas, Xpm requiere efectores tipo III (T3E) para su plena virulencia, colonización bacteriana y el desarrollo exitoso de la CBB. XopAE, un efector de la familia de proteínas externas de Xanthomonas (Xop), constituye uno de los T3E más conservados en Xpm. Este trabajo tuvo como objetivo evaluar la interacción molecular entre XopAEXpm y las patelinas MePATL3 y AtPATL3. Nuestro análisis bioinformático sugiere que MePATL3 es una proteína de tipo Sec14-GOLD PITP, y que presenta homología estructural con AtPATL3 basado en la identificación de dominios CRAL/TRIO y GOLD altamente conservados. Además, realizamos ensayos de doble híbrido de levadura (Y2H, por sus siglas en inglés) para evaluar la interacción entre XopAEXpm y MePATL3 en su extensión completa, así como entre XopAEXpm y versiones truncadas de AtPATL3, lo que nos permitió identificar la región N-terminal y/o el dominio C-terminal CRAL/TRIO de AtPATL3 como puntos críticos de interacción con XopAEXpm. Por último, realizamos ensayos de Western Blot expresando MePATL3 en N. benthamiana como un primer paso para los ensayos de Co-Inmunoprecipitación in planta.

    Laying the groundwork for protein-protein interaction experiments for Toxoplasma gondii Dihydroorotate Dehydrogenase: preparation and preliminary design

    Toxoplasma gondii is an intracellular parasite of clinical and veterinary significance, capable of infecting a wide range of hosts. Despite the availability of treatments, challenges such as chronic infection, drug efficiency, and strain-specific drug resistance persist. Targeting nucleotide metabolism, particularly the pyrimidine synthesis pathway, has shown promise in related parasites. Dihydroorotate Dehydrogenase (DHODH), a crucial enzyme involved in T. gondii's pyrimidine metabolism, has been identified as a potential drug target in related parasites, and previous knockout assays in T. gondii revealed that DHODH holds an essential non-pyrimidine-related role that remains uncharacterized. This project aims to perform the first steps to identify protein-protein interactions involving TgDHODH, and the specific domains mediating these interactions through blue native electrophoresis, pull-down assays, and yeast two-hybrid assays. First, the N-terminal domain of TgDHODH (NT-TgDHODH) was successfully cloned into the pET15b expression vector and expressed in E. coli. Purification of the recombinant protein presented challenges due to low protein yield and insolubility. Secondly, BN-PAGE coupled with DHODH activity assays revealed the presence of TgDHODH in two bands migrating at higher molecular weights than expected for TgDHODH alone. These bands were excised and will be subjected to mass spectrometry analysis to identify potential protein partners. Finally, the cloning of TgDHODH and NT-TgDHODH into the pGBKT7 bait plasmid for the yeast two-hybrid assay was successfully accomplished. The vectors containing the bait proteins were stored for future use in subsequent Y2H assays. Although progress has been made in this project, further experiments are required to identify protein partners and characterize the unknown, pyrimidine-independent function of TgDHODH. The purification of NT-TgDHODH, along with pull-down assays and mass spectrometry analysis, will provide valuable insights into the protein-protein interactions of the enzyme. Furthermore, the subsequent yeast two-hybrid assay will confirm the identified interactions. Understanding the function and protein interactions of TgDHODH will enhance our knowledge of the parasite's biology and facilitate the development of new drugs against toxoplasmosis.

    Evaluación del efecto aditivo de bacteriófagos contra Cutibacterium acnes e isotretinoína en un modelo de cultivo celular

    El acné vulgaris es una enfermedad inflamatoria crónica de la piel, la cual afecta en mayor medida a la población entre 12 y 24 años, aunque puede llegar a afectar a personas de hasta 45 o más años de edad. Esta enfermedad tiene una alta prevalencia, causando diferentes problemas psicológicos y sociales. Actualmente, existen diferentes tratamientos tópicos y orales como los retinoides, los antibióticos, la isotretinoína, entre otros; los cuales han demostrado tener una gran eficacia en el control del acné. No obstante, se ha demostrado que estos presentan desventajas importantes como la aparición de resistencia a los antibióticos por parte del patógeno Cutibacterium acnes y la aparición de efectos secundarios en diferentes sistemas del organismo. Por ende, es necesario la búsqueda de nuevos tratamientos. Una de estas alternativas es el uso de bacteriófagos, los cuales causan la lisis celular del patógeno, reduciendo así su abundancia en los folículos pilosos. No obstante, no logran actuar sobre los otros factores etiológicos del acné. Por lo tanto, en esta investigación se propone generar una nueva alternativa, al usar bacteriófagos con moléculas de isotretinoína unidas a su superficie, con el propósito de generar un tratamiento localizado y dirigido, para reducir los efectos secundarios del medicamento, el cual contaría con actividad antiinflamatoria (isotretinoína) y antimicrobiana (bacteriófagos). Para esto, se emplearon una cepa de C. acnes denominada Pa6 y el bacteriófago PAc2, los cuales fueron aislados en investigaciones previas de pacientes con lesiones de acné. Asimismo, se llevaron a cabo diferentes procesos para seleccionar los blancos y lograr la unión estable de las moléculas de isotretinoína a las proteínas de cápside del bacteriófago, se realizaron ensayos de inocuidad del estos fagos por medio de pruebas de citotoxicidad in vitro en cultivos celulares de queratinocitos y se evaluó la actividad antiinflamatoria por medio de la cuantificación de los niveles de expresión de la IL-1B . Los resultados del análisis bioinformático mostraron que es posible realizar la unión por medio de la formación de enlaces ésteres, gracias al extremo carboxilo del fármaco y a los residuos hidroxilo de las proteínas estructurales del bacteriófago , lo cual se corroboró de manera experimental por medio de espectrofotometría UV (Información que se encuentra en proceso de protección de propiedad intelectual). De igual forma, se determinó que aunque el título viral del bacteriófago decorado disminuye, cambiando de 1.1*1011 UFP/mL a 2.2*105 UFP/mL, este sigue teniendo la capacidad de infectar a la bacteria. En cuanto a los ensayos de inocuidad en el cultivo celular, se realizaron pruebas con el medicamento y el bacteriófago de manera individual y en la formación del complejo, los cuales mostraron que no se afecta la viabilidad de la línea celular. Asimismo, el tratamiento propuesto logra reducir los niveles de expresión de la interloquina proinflamatoria. En conclusión, esta aproximación puede llegar a ser una gran alternativa a las terapias actuales contra el acné, ya que puede llegar a actuar sobre varios de los factores fisiopatológicos que causan la enfermedad y puede generar mayor seguridad para el paciente al reducir los efectos secundarios.

    Actualización del conocimiento sobre taxonomía y distribución de los satúrnidos de Colombia (Lepidoptera: Saturniidae)

    En los últimos años, el estudio de los Saturniidae ha aumentado exponencialmente gracias al intenso esfuerzo de muestreo y al uso de evidencia molecular para la delimitación de especies, lo que ha llevado a la descripción de numerosos taxones nuevos. Dados estos rápidos avances, el listado de especies de Saturniidae de Colombia se encuentra desactualizado, por lo que se requiere un trabajo sistemático que aborde la taxonomía, distribución y diversidad de estas polillas en el país, además de actualizar las claves de identificación existentes. El objetivo de este trabajo es abordar estos vacíos de información producto de los avances recientes y realizar un análisis biogeográfico y altitudinal de los Saturniidae en Colombia. Se llevó a cabo una exhaustiva revisión de literatura y colecciones biológicas, junto con muestreos adicionales en 70 localidades previamente no exploradas. Se elaboró un mapa de regiones biogeográficas considerando la distribución de los taxones y los patrones de endemismos observados. Como resultado, se presenta una revisión taxonómica y una discusión biogeográfica de los Saturniidae en Colombia, que incluye una clave dicotómica para las 7 subfamilias presentes en el país, un listado nacional actualizado y el reporte de 51 nuevos registros de especies para el país. Colombia destaca como un país con una notable diversidad de Saturniidae, con un total de 732 especies y subespecies en 55 géneros, siendo además altamente endémica, con 328 especies y 14 subespecies exclusivas del territorio colombiano, debido a su compleja geografía. En este estudio se han resuelto confusiones taxonómicas, corrigiendo el uso de nombres científicos desactualizados y proponiendo cambios taxonómicos necesarios. A nivel geográfico, se destaca la importancia de las Cordilleras andinas y la Sierra Nevada de Santa Marta, así como de los bosques altoandinos y los páramos, como áreas especialmente relevantes para la conservación de esta familia de polillas, debido a los endemismos que encierran. Este estudio proporciona una valiosa herramienta para la investigación y conservación de los Saturniidae en Colombia, así como para futuros estudios de biodiversidad y biogeografía en la región neotropical.

    Rol del T6SS en Competencia Bacteriana de Xanthomonas phaseoli pv. manihotis Ante Bacterias Epífitas de Yuca (Manihot esculenta)

    Mediante el Sistema de secreción tipo VI (T6SS), las bacterias gramnegativas pueden translocar proteínas efectoras que juegan un papel importante en competencia bacteriana y virulencia hacia hospederos eucariotas. En este estudio, fue posible realizar el aislamiento e identificación taxonómica mediante secuenciación del gen 16S, de seis bacterias epífitas que habitan la filósfera de yuca (Manihot esculenta), una especie vegetal de importancia económica en Sur América y África. Ensayos de competencia bacteriana in vitro sugirieron que el T6SS de la bacteria fitopatógena Xanthomonas phaseoli pv. manihotis (Xpm) era requerido por contacto directo para ejercer competencia bacteriana hacia la bacteria epífita de yuca Rhodococcus sp. Mutantes para los genes VgrG, ClpV y Hcp del T6SS de Xpm fueron incapaces de inhibir el crecimiento de Rhodococcus sp. Sin embargo, no fue posible determinar con certeza que el T6SS de Xpm sea el responsable de la inhibición de Rhodococcus sp., debido a la falta de complementación observada de estos mutantes en Xpm. Finalmente, fue posible predecir a través de análisis bioinformáticos dos proteínas efectoras que serían hipotéticamente traslocadas por el T6SS de Xpm, y el papel que podrían cumplir en la competencia bacteriana. Los resultados obtenidos en este estudio ayudan a comprender de alguna manera, cómo el T6SS podría jugar un papel importante en competencia bacteriana posibilitando que Xpm gane ventaja de nicho al matar a otras bacterias, facilitando finalmente el establecimiento de la enfermedad tizón bacteriano de la yuca (CBB).

    The Quest for the Essentials: Analyzing Microbial Successional Diversity in a Novel Experimental Material at Two Island Locations in Colombia

    The microbial successional process has been studied with different materials to assess biodiversity and its relationship with other environmental variables. Nevertheless, it has been shown that the material used and its position directly impact the type of microorganism that will attach. Therefore, the use of a standardized material and method is a necessity. Here, we evaluate the microbial successional diversity on San Andrés and Gorgona Island, Colombia, and its relationship with depth. Experimental blocks were placed on two different diving depths (6 and 12 meters) for nine months. Every two months, blocks were collected for each study site and the two different depths. DNA extraction and PCR amplification were performed on each block. After the end of the experiment, all DNA isolations were sent for library preparation and sequencing for bacteria, fungi, and eukaryote identification with their corresponding marker genes 16sV4, ITS2, and 18sV9, respectively. Taxonomic identification was performed using QIIME2 and analyzed with alpha and beta diversity metrics and a differential abundance analysis. Our results indicate that different taxonomic groups vary in their alpha and diversity metrics based on their study site and depth. Proteobacteria was more abundant in San Andrés samples and Cyanobacteria in Gorgona samples. On eukaryotes, Annelida and Arthropoda were the most abundant in both study sites. Lastly, on fungi, Ascomycota was the most abundant phylum. In conclusion, the experimental units attracted many microorganisms to dwell and start their natural successional processes. Using a standardized method allowed us to compare study sites and variables, such as depth.
