Diseño de Control para un Toggle Switch alrededor de su inestabilidad

El Toggle Switch es un circuito genético que tiene dos genes que se reprimen entre sí. Este sistema biestable, también permite regular la represión mediante la concentración de las moléculas inductoras asociadas al medio de crecimiento. Se estudia la dinámica del sistema de un Toggle Switch particular(capítulo 3), cuyo modelo surge a partir de un ajuste a datos obtenidos experimentalmente, y será la base para el estudio en este trabajo. Además, se caracteriza el sistema del modelo determinístico obteniendo sus puntos de equilibrio, dos estables y uno inestable, donde las proteínas tienen una producción relativamente baja. El objetivo de este trabajo es mantener el sistema alrededor de estos estados intermedios antes de la diferenciación. La respuesta del modelo determinista podrá corresponder al promedio de una población, la naturaleza de la expresión génica es estocástica, de ahí el interés de tener un modelo que represente un organismo en particular. Es así como en el capítulo 4, se hace un modelo de la transcripción y traducción de un gen constitutivo en sus respectivos procesos para la creación y destrucción; primero, a través de la Ecuación Maestra para obtener distribuciones más exactas pero con un alto costo analítico, y posteriormente, desde la Ecuación de Langevin siendo un modelo más intuitivo desde lo fenomenológico, pero a costa de exactitud. De allí, observar la respuesta del sistema y diversas características como el tiempo de establecimiento, la respuesta acotada o no del sistema al considerar la estocasticidad, entre otros. Finalmente, se propone un diseño de un control lineal por retroalimentación negativa sobre el modelo determinista, para mantener al sistema en estados 'cercanos' del punto inestable. El diseño del control se realiza para unas especificaciones dadas, tales como una respuesta oscilatoria determinada y un tiempo de establecimiento del mismo orden de magnitud a la respuesta obtenida en la caracterización del sistema.

The Toggle Switch is a genetic circuit that has two genes that repress each other. This bistable system also allows regular repression through the concentration of the inducer molecules, which are associated with the growth medium. The dynamics of the system of a particular Toggle Switch (Chapter 3) is studied, whose model arises from an adjustment to experimentally obtained data, and will be the basis for the study in this work. Besides, the deterministic model system is characterized by obtaining its equilibrium points, two are stables ant the other is unstable, where the proteins have a relatively low production. The objective of this work is to maintain the system around these intermediate states before of differentiation. The response of deterministic model could be the average of population, the nature of gene expression is stochastic, hence the interest in building a model that represents a particular organism. Then in chapter 4, a model is made of the transcription and translation of a constitutive gene in their respective processes for the creation and destruction, first through the Master Equation to obtain distributions more exact but with a high analytical cost, and later, from the Langevin equation being a more intuitive model from the phenomenological, but at the cost of accuracy. So, the response of the system and various features such as the establishment time and the bounded response of the system are observed when considering stochasticity. Finally, a design of a linear control by negative feedback on the deterministic model is proposed, to keep the system 'close' the unstable point. The control design is carried out for given specifications such as a determined oscillatory response and a settling time of the same order of magnitude as the response time obtained in the characterization of the system.
